Biocompatibility evaluation of common used mouthwashes

Abstract (EN)

The aim of this study is to evaluate the effects of 6 commonly used mouthwashes on the viability of L929 mouse fibroblast cell with XTT and real-time cell analysis methods. For the xtt assay, %5, %10 and % 20 concentrations of Kloroben, Curasept ADS 205, Curasept ADS 212, Curasept ADS 220, Klorhex and Andorex material extracts were prepared. L929 cells were exposed to material extracts for 1 minute, 5 minutes, and 24 hours 2 times per day (n=12) . Data were statistically evaluated with one-way ANOVA analysis. For real-time cell analysis assay, culture medium was added to the control wells to obtain the maximum dose of the extract suspension, while the others had a 5% concentration (n=12).E-plates were measured every 15 minutes for 125 hours. Results were analyzed by RTCA 2.0 program. According to XTT test data, the effect of all concentrations of materials on the viability of L929 cells at different application were different from the control group (p<0.05). According to the results of real-time cell analysis, it was observed that cell viability values of Kloroben, Curasept ADS 205, Curasept ADS 212, Klorhex and Andorex were very close to 0 after 125 hours and for Curasept 220 it was below. Mouthwashes may cause cytotoxic effects in the mouth due to their biologically active content. In this study, data were obtained by performing 2D in vitro cell culture tests. 3D cell culture tests and in vivo studies should be performed to obtain data closer to the clinic.
