The protective effect of N-acetylcysteine in reversalof sulfasalazine related renal and testicular toxicity

Abstract (EN)
The toxic affects of sulfasalazine (SASP), which has been commonly used for the rheumatic disease, the treatment of ulserative colitis and chron disease for a long time, and to what extent these affects have been able to be reversed by N-Asetylcysteine on the Spraque-Dawley type of rats in suitable laboratory setting have been studied. Spraque Dawley types, 28 male rats weighting 220-369 gr, have been homogenised according to their rat weight and they are divided into four groups that each group has seven rats. Three of these groups have been determined as medication and the other one has been as control group. Suffasaline 600 mg/kg and N-acetylcysteine 150 mg/kg, which have been dissoved in water, have been given to one of the medication group by gavage cannula one in every 24 hours. This process has been carried out for 14 days. The tissues of rats have been taken out with general anesthesia without sacrificed. Tissue samples have been seperated for pathological and biochemical examination after being weighted. In biochemical examination, MDA values in tissue and the activities of glutathione, catalase and superoxide dismutase have been evaluated. The pathological examination has been done by Olympus BX51 light microscopy. It has shown that sulfasalazine raises the MDA values in testiculary and renal, and also it pathologically ruins the sperm quality in testicular and increases the tübülery degeneration in renal. In this study, it is proved that N-Asetylcysteine has a protective affect in sulfasalazine toxicity by decreasing the MDA value and tübüler degeneration in renal and also by increasing the sperm quality in testicular.
