Investigation of interference effect of hemolysis in tbars-malondialdehyde (MDA) analysis

Abstract (EN)

Hemolysis is passing of all of the material of erythrocyte membrane, especially hemoglobin, into the serum or plasma as result of erythrocyte membrane damage. It has been shown that when the hemolysis is composed of nearly 15-20 mg/dL of free hemoglobin concentration, hemolysis started to interfere blood parameter. Hemolytic interference, an analyte in serum or plasma depends on effects of hemolysis, is involuntry change which is generated in analyte concentration. Especially spectral interference is one of the most important of them. Hemolysis causes to spectral interference especially in 540 and 570 nanometer. Malondialdehyde is a lipid peroxydation assessment method is used mostly in basic research and spectrometric measuring is done in 532 nm. The aim of this study: investigation of interference effects of hemolysis on MDA analysis and identifying of threshold value for hemolysis which affect MDA measurement. As a result both MDA's serum pools (low and high) that is still studied intensely, it was observed that MDA stages were effected. Hemolysis index caused deflection at approximately between intervals of 400-450 at high MDA serum stages, this vaule caused deflection at approximately between intervals of 500-600 at low MDA serum stages. Therefore these values and above of them shouldn't be included to studies because they need tolerance. Also it was observed that there were higher interferences at low serum MDA groups.
