Abstract (EN)
Because of its broad substrate specifity, the laccase enzyme can be used in many industrial fields.Although the white rot fungi are the best laccase producer organisms, the amount they produce is not sufficient for industrial use. Hence, it is necessary to optimize the conditions in order to increase laccase enzyme production to be used in different fields. Funalia trogii and Trametes versicolor which are good laccase producers were used in this study and various culture conditions were optimized.The optimization works were conducted in ?repeated batch process?, which proved more advantegous to ?batch process?. Most of the the studies were performed in SBM (Stock Basal Medium) and SBM+0.5 mM Cu media. The effect of agitation, initial pH, incubation temperature, amount of pellet, time interval and copper concentration on laccase production was investigated and optimum laccase production conditions were determined for both fungi. After that, the effect of some inducers such as ABTS [2,2'-Azino-bis(3-ethylbenzthiazoline-6-sulfonic acid)], syringaldazine, guaiacol and 2,5-xylidine on laccase production ability of these fungi was tested under optimal conditions and the most effective inducers and their concentrations were determinated for both fungi.In the last stage of the study the molecular weights of the laccases, which were synthesized by F. trogii and T. versicolor, were determinated by SDS PAGE.The results of this study show that it is possible to obtain high amounts of laccase by using the suitable fungi, processes and inducers under optimal conditions.
