Microbial identification with the vibrational spectroscopy

Abstract (EN)

In this study, identification of eukaryotic microorganisms was aimed by using vibrational spectroscopy. Two methods of vibrational spectroscopy are Infrared and Raman. Although these two methods are connected to each other, the infrared (IR) and Raman spectra are not exactly the same and, each has its own characteristics. Five species of yeasts and three species of dermophytes were grown on SDA and their profiles were analyzed by FT-IR and Raman spectroscopy. The signal enhancement was observed via SERS, which is the method of Raman spectroscopy, and the spectra obtained was compared using Hierarchical Cluster Analysis by SPSS statistics programme. It was observed that yeasts; Candida albicans, Candida tropicalis, Candida krusei, Candida parapsilosis, Trichosporon asahii congregated in one cluster and the dermatophytes; Microsporum canis, Trichophyton rubrum, Trichophyton mentagrophytes congregated in another cluster. It was concluded that, the silver nanoparticles and the strength of the interaction between the cell walls of the microorganisms depend on the biochemical structure by looking at the images taken from scanning electron microscope (SEM).
