Identification and sequence analysis of yersinia ruckeri infection in trout in Tunceli province by maldi-TOF MS

Abstract (EN)

It is an Enteric Scarlet Mouth Disease caused by Yersinia ruckeri, which is commonly seen in rainbow trout in our country. Yersinia ruckeri, a member of the Enterobacteriaceae family, is an injection-causing pathogen in humans and fish. This disease causes high mortality, especially in juvenile trout. Epidemics are widely observed in fish farms all over the world and in our country and lead to economic losses. The aim of this study is; April May and June in the province of Tunceli, Mazgirt, Çemişgezek, Ovacık and Pertek districts in the network cages and concrete pools of trout farming enterprises that show signs of disease from 200 juvenile or adult rainbow trout and pool water samples containing trout to detect the presence of Yersinia rucker by conventional methods, collecting data on the prevalence and distribution of this factor in the region, identification of the factor with the MALDI-TOF MS method, to compare the MALDI-TOF MS and 16S rDNA sequence analysis methods used in the identification of Yersinia ruckeri. As a result of the analysis, 401 (66.83%) bacteria belonging to 9 family groups, including Enterobacteriaceae, Enterococcaceae, Moraxellaceae, Bacillaceae, Aeromonadaceae, Staphylococcaceae, Streptococcaceae, Comamonadaceae, Pseudomonadaceae, were identified. Yersinia ruckeri was detected with MALDI-TOF MS at a rate of 0.5% (3/600) from the facilities in Tunceli province and districts. A specific primer was used to confirm the isolates identified as Yersinia ruckeri by PCR technique. Its accuracy has been proven by undergoing 16S rDNA sequence analysis and obtaining successful results. Yersinia ruckeri could not be detected in the water samples. As a result; in this study, the diagnosis of Yersinia ruckeri was accurately and definitively identified by the MALDI-TOF MS method in rainbow trout in the province of Tunceli. In this respect, our study is the first study conducted in the province of Tunceli, located in the Upper Euphrates Basin of the Eastern Anatolia Region. This study will contribute scientifically and will be a guide for further studies The MALDI-TOF MS method, which is used for rapid and reliable identification of bacteria, will prevent the spread of infectious diseases and economic losses in aquaculture. Identification of the causative agent in fish diseases will contribute to the development of effective treatment and eradication strategies.
