Abstract (EN)

The aim of this study was to investigate the hydrolytic enzyme potentials of halophilic microorganisms isolated from salt and ponds such as Bingöl, Kemah, Fadlum, Sivas. A total of 188 isolates were incubated with nuclease screening in DNase test agar medium, 2% milk powder in the protease screening medium and incubation with medium containing amylase 0.5% starch at 37 ° C for 24 hours. Enzyme production potentials were determined according to hydrolytic zone diameters around the colony. DNA isolation and 16srRNA PCR for unspecified isolates were established and 7 different patterns were obtained by ARDRA and the next step was prepared for sequence analysis studies. D85, 205y, F2S50 and D63 microorganisms were determined as microorganisms with the highest enzyme activity according to the zone diameters. The growth curves of these microorganisms and the highest protease enzyme activity hours were determined. In future studies, the identification of unidentified isolates by sequence analysis, the peak activity of amylase and nuclease enzymes are determined and largescale industrial production is targeted.
