Abstract (EN)

The aim of this study was to determine the effects of bacteria obtained from vermicompost on the bacterial canker and wilt disease (Clavibacter michiganensis subsp. michiganensis. As a result of the isolations, 40 isolates were obtained from vermicomposts. Pectolytic activity test on potato, hypersensitive reaction test on tobacco, growth test at 37 °C were performed with the isolated isolates and phosphorus reducing and nitrogen solubilizing properties of the isolates were determined. A dual culture test was performed with 22 selected isolates and pathogen bacteria C. michiganensis subsp. michiganensis and 10 bacterial isolates were effective on the pathogenic bacteria at rates ranging from 2.45 to 11.98 mm. Seed study was carried out with 3 bacterial isolates (SA-2, SA-5, SA-7) selected because of these tests. Bacterial isolates were applied to pathogen-contamined seeds as coating and sown in vials under greenhouse conditions. Selected bacterial isolates were applied to the seeds contaminated with the pathogen as coating and sown in vials under greenhouse conditions. In the evaluations made by considering only the seedling development in the seeds applied with the pathogen, the highest effect in suppressing the disease was seen in the SA-7 (Bacillus megaterium) isolates with 70%. Following this isolate, the Bacillus sp. isolate with code SA-5 also showed 64% efficacy. In addition, the effect of isolates on seedling development was also investigated in the study. According to the materials and methods used, no plant growth enhancing effect of bacteria was observed in seedlings developed from seeds to which only selected bacterial isolates were applied and no pathogen applied. This master's thesis study aims to determine the mechanisms of actions Bacillus megaterium SA-7 and Bacillus sp. SA-5 isolates effective in seed treatment and to demonstrate their pathogen effectiveness through greenhouse and field studies. The results will provide a foundation for future studies.
