Abstract (EN)

Within the scope of the thesis, antibiotic-loaded alginate-gelatin-hyaluronic acid-based hydrogels were developed to be used as a wound dressing material. In the first step, alginate, gelatin and hyaluronic acid methacrylation reactions were performed and the obtained products were characterized by Fourier transform infrared (FTIR) and proton nuclear magnetic resonance (1H-NMR) spectroscopy. In the second stage, alginate methacrylate-gelatin methacrylate (AG), alginate methacrylate-hyaluronic acid methacrylate (AH) and alginate methacrylate-gelatin methacrylate-hyaluronic acid methacrylate (AGH) hydrogel formulations were developed. Hydrogel structures were examined by FTIR and scanning electron microscopy (SEM). In addition, water holding capacity of hydrogels and in vitro hydrolytic degradation studies were carried out. Next, tetracycline-loaded hydrogels were prepared and characterized by FTIR and differential scanning calorimetry (DSC). In vitro release studies were performed at 37 °C and in PBS medium. Its antibacterial properties were evaluated against S. Aureus, E. Coli and S. Epidermidis bacteria using the disc diffusion method. Biocompatibility studies were determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyl tetrazolium bromide (MTT) method performed using the L929 cell line. All results showed that the developed tetracycline-containing hydrogel dressing material has promising potential. Key Terms: Hydrogel, wound dressing material, antibiotic release
