The effect of LET7-B, MIR29-A levels on treatment success in assisted reproduction applications

Abstract (EN)

Aim: Infertility has become the plague of our time, reproductive disorders and sometimes the difficulty of treatment processes bring a very heavy financial and moral burden on couples. In this study, it was aimed to observe the differences in the expression and expression levels of the two microRNAs between the two groups we created after treatment, and to investigate the effects of serum levels of Dnmt3a and Erα associated with these two microRNAs we selected. Materials and Methods: A total of 60 infertile patients who applied to Ondokuz Mayıs University Hospital IVF treatment center participated in our study. RNAs were purified from peripheral blood samples using standard methods. Quantitative simultaneous PCR technique was used to measure and compare the expression and expression levels of mir-Let7b and mir-29a. Dnmt3a and ERα levels were measured by ELISA technique. The data were evaluated statistically. Results: The differences in miR-Let7b, miR-29a gene expression and expression levels of 60 infertile patients who applied to the OMU hospital in vitro fertilization treatment center were significantly different between the group that became pregnant after the treatment and succeeded and the two groups that failed to conceive (p=0.04; p=0.03), respectively. In addition, differences in the concentration and expression levels of the two serum Dnmt3a and ERα associated with these two microRNAs we selected later were also detected between the two groups. It was determined that there were significant differences between the two groups, respectively (p=0.04 ; p=0.03). , for ERα (r=0.541) and (r=0.508), respectively, for Dnmt3a there was a negative correlation from age (r = -0.654) and a positive correlation from body mass index (r = 0.542) (p<0.05). Conclusion: In our study, it has been shown that the difference in microRNA expression and expression levels between the two groups of infertile patients has an effect and role on the patients in the post-treatment period. Our study is the first study in our country in which the effects of miR-Let7b and miR-29a expression levels on the process of achieving success after treatment in infertility patients were investigated.
