Abstract (EN)

Since the liver is responsible for metabolizing many of the drugs, including antiepileptics, the main target of drug-induced damage is this organ. Carbamazepine which is widely used in the liver and metabolized, is a prescribed drug, despite its potential for hepatotoxicity. On the other hand, perampanel is a new antiepileptic that has been approved in 2012 and underwent extensive hepatic biotransformation. There is very little data on perampanel, which is an important class in terms of risk of developing hepatotoxicity, yet related to its use. Therefore, it was aimed to determine the hepatotoxicity of perampanel using HepG2 cell line in vitro compared to carbamazepine which has hepatotoxicity potential. HepG2 cytotoxicity on the HepG2 cell line used, hepatotoxicity was calculated by MTT cytotoxicity test inhibitor concentrations, followed by determination of apoptosis / necrosis, various biochemical analyzes (ALT, AST, urea) and ROT determination which are currently a biomarker for liver damage. According to the results, it was observed that perampanel, which was studied in the thesis, showed cytotoxic effect in the HepG2 cell line like carbamazepine, caused an increase in hepatic biomarkers and induced oxidative stress.
