Özet (EN)
Cancer is still a very significant health problem and there is still not enough success its treatment despite new drugs. Therefore, the search for a new compound in cancer therapy is going on. The aim of this study is to determine the anticancer activity of novel Pd (II) complex on transformed fibroblast cells. For this purpose, MTT and ATP cell viability tests are utilized to determine in vitro cytotoxicity. Furthermore, cytotoxic activity is confirmed with real time cytotoxicity analysis system (xCELLigence RTCA). Flow cytometry is used to determine the cell death mechanism (apoptosis/necrosis) in cytotoxic/cytostatic effects of Pd (II) complex on fibrosarcoma cells. Presence of apoptosis is supported with Hoechst 33342/PI/Calcein-AM triple staining method in fluorescence microscope. In conclusion, it has been determined that novel Pd (II) complex has an antiproliferative effect on fibrosarcoma cells by inducing apoptosis. It has been concluded that there is a need for in vivo experiments with the prediction of usage of novel Pd (II) complex as a therapy in human fibrosarcomas. Keywords: Cancer, Palladium (II) complex, Sarcoma, Apoptosis
