Abstract (EN)
Many of the traditional chemothrapeutic agents used in the treatment of breast cancer(BC) cause systemic toxicity. For this purpose, studies conducted with drug candidate molecules called new generation with low cytotoxic effects and high anti-cancer effects have gained importance in recent years. Amino anthroquinones (AA) and it derivatives are widely used for the cancer treatment today. One of the most common examples of AA used in BC standard therapy, mitoxantrone (MTX) is included in the cytotoxic antibiotic class. In this thesis study, the synergistic effect of the newly synthesized AA-derived molecule on human BC cells (MCF-7) and Human Umbilical Vein Endothelial Cells (HUVEC) . For this hence In this direction, cytotoxicity tests were carried out by MTT method and the obtained data were analyzed and the inhibition concentrations IC50 at which the cells were 50% viable were calculated.Apoptotic analyzes were performed using flow cytometry method. During the experiments, AA and MTX were encapsulated individually together at appropriate concentrations in order to maximize bioavailability. Cytotoxicity tests and apoptotic analyzes were performed by repeating all experiments with these derivatives and drug combinations were encapsulated . At the end of 48 hours, it was observed that their effectiveness increased compared to the unencapsulated forms. In the results we obtained, the cytotoxic effect of AA in HUVEC cells used as control highlights the synergistic effect of the drug. We anticipate that all these studies will form the basis for future in vivo experiments and will make an significent contribution to the development of new drug candidate molecules in BC treatment.
