Abstract (EN)

Most women are diagnosed with ovarian cancer at an advanced stage, as it is asymptomatic at an early stage and lack of adequate early detection screening methods. Molecular pathogenesis of the disease should be explained for early diagnosis, treatment and follow-up of ovarian cancer. In this way, new biological markers are needed. MicroRNAs are a class of small non-coding 18-25 nucleotides in length RNA molecules that regulate gene expression at the posttranscriptional level. It has been shown to have a very important function in oncogenesis by regulating cell proliferation, cell differentiation, and apoptosis as an oncogene or tumor suppressor. A large number of miRNAs have been shown to be associated with ovarian tumor genesis, and some of these miRNA genes have been shown to be ideal targets for diagnosis, prognosis, and treatment. In the thesis study presented, the expression level of miR-27a-3p selected among miRNAs previously determined by our group to be important for ovarian cancer was investigated in the peripheral blood of ovarian cancer patients and healthy individuals. Expression analyzes using RT-PCR technique were statistically evaluated using SPSS v21.0 program. As a result of Mann-WhitneyU test and ROC analysis, it was determined that miRNA, which was studied between healthy controls and ovarian cancer patient groups, caused a significant difference. In ovarian cancer patients, miR-27a-3p expression was shown to increase by 1.95 times compared to the control group, and this increase was calculated to be statistically significant (p <0.05). In the literature, it has been reported that miR-27a-3p is over-expressed in tumor tissue of many cancers, including ovarian cancer, but there is no information about the level of miR-27a-3p in the peripheral circulation of cases with ovarian cancer. By the presented thesis study, it was determined for the first time that the cases with ovarian cancer increased the level of miR-27a-3p expression in the peripheral circulation, and that the molecule in question has the potential to be a biological marker. In future studies, it is thought that the prognostic significance can be demonstrated by using the miR-27a-3p expression level in the diagnosis and treatment of the disease.
