Thesis, Istanbul. In this study, ten novel compounds having 1-[4-hydroxy-3-[(substituted-1- piperazinyl)methyl]phenyl]ethanone backbone were prepared by Mannich reaction using conventional and microwave assisted synthesis methods. To obtain the target compounds in conventional method a mixture of 4-hydroxyacetophenone, formaldehyde and suitable piperazine derivative were reacted in methanol for 48 hours under reflux at 65 °C. For the microwave assisted synthesis a mixture of 4-hydroxyacetophenone, formaldehyde and suitable piperazine derivative were reacted in 1,4-dioxane for 30 minutes at 120 °C. A comparison between the two methods were carried out yield wise. Structure elucidation of the synthesized compounds were confirmed by IR, 1H NMR, 13C-NMR, and elemental analysis. The cytotoxicity of the compounds was examined against human breast cancer cell line (MCF-7) by MTT assay. According to the activity data, compounds 4 and 7 had the highest cytotoxic effect.
