Detection and molecular characterisation of tomato chlorosis virus (Tomato chlorosis crinivirus, tocv) in Adana and Mersin

Abstract (EN)
Tomato Chlorosis Virus (ToCV) is a member of the genus Crinivirus of Closteroviridae family and important virus on tomato. ToC has large host range including ower 24 plant species in 7 plant families. The research was conducted to disply prevalence of virus, classification of different isolates on tomato produced in Adana, Mersin provinces. Due to lack of antiserum of ToCV commerciall, no possibility to detect of ToCV serologically. During surveys, the samples were tested with ELISA against to Cucumber Mosaic Virus (CMV) and Tobacco Mosaic Virus (TMV) to control of mix infection of tomato. The samples found infected with ToCV by molecular techniques were inoculated to tomato, pepper and indicator plants by mechanically inoculation technique. Biological indexing showed that ToCV is not transmited by mechanically inoculation. Different isolates of ToCV wre characterized by RT-PCR according to 360 bp fragment on coat protein gene. RT-PCR assays showed that the agent of symptoms of chloroz between veins, yellowing, browning and flecking is ToCV. When sequences of ToCV isolates found were compared with other isolates in genebank, the result showed that our isolates are very close to Greec isolates in that rate of 98,9%.
