ABSTRACT Master Thesis RESEARCH OF MELATONIN AND VITAMIN E-SELENIUM MIXTURE EFFECTS TO CREATED CHANGE OF CADMIUM ON FATTY ACH) COMPOSITION IN ALBINO RATS Mahmut YILMAZ Firat University Graduate School of Natural and Applied Sciences Department of Biology 2005, Page: 38 The objective of the work which was carried out in this study was to investigate the influences of antioxidant melatonin and antioxidant Vitamin E- selenium to the variation of fatty acid compounds on the rats liver, kidney, heart, testis tissues with chronic cadmium effects. The rats (specimens) which were treated with cadmium (Cd), melatonin and vitamin E+selenium for a period of one month. They were divided into five groups consisted of 10 rats; 1) a control group 2) cadmium group 3) cadmium+ melatonin group 4) cadmium + vitamin E+selenium group and 5) cadmium+melatonin+vitamin-E+selenium group. Throughout one month the Rats in cadmium group were given 1 mg/kg CdCl2, 10 mg/kg melatonin, 30 mg/kg+2 ug Vitamin-E+selenium as overdose. At the end of the experimental period the specimens (rats) in five groups were sacrified after that the variation of fatty acid compounds on their liver, kidney, testis and heart tissues were analyzed with gas cromatography. The fatty acids such as palmitic acid (C16:0), stearic acid (C18:0), oleic acid (C18:l), linoleic acid (C18:2n6), arachidonic acid (C20:4) and dokosahekzaenoic acid (C22:6) were determined on the rats tissues (liver, kidney, heart, testis). For the groups treated Cd, the level of fatty acid (CI 8:1 n7) on the liver tissue was lower (PO.05) than control group, but it was significantly higher than testis groups. The fatty acids of CI 8:1 n9 and C20:4n6 were higher (P<0.01) than control groups treated Cd on the testis and kidney tissues. It was determined that there was a negative correlation (r = -0.869) between C18:2n6 and C20:4n6 on the kidney tissue. A negative correlation (r = -0.904) was determined between C18:ln9 and C20:4n6 on the testis tissues. In conclusion, our findings showed that the exposure of heavy metals such as Cd have inhibitor effects on enzimes charged for A-9 desaturase and A-6 desaturation steps as a result of their reduced effects on unsaturated fatty acid synthesis.
