Investigation of the common candidare molecular biomarkers and key pathways that play a role in the pathogenesis of alzheimer's disease and cancer

Abstract (EN)

In recent studies, a link has been established between Alzheimer's Disease and cancer, but the common mechanism has not been adequately investigated. It has been known that biological pathways such as MAPK/ERK, Wnt and PI3K/Akt /mTOR play a role in the shared etiology yet it is important to detect common biological processes and molecular markers associated with two diseases to establish new translational approaches. In this study, the relationship between AH and 4 cancer types (breast, colorectal, non-small cell lung cancer, prostate) was comprehensively investigated by analyzing microarray data using bioinformatics tools in order to enlighten the common molecular pathways and biomarkers. Microarray datasets were selected according to the certain criteria in NCBI-GEO database and analyzed by using R software in GEO2R platform. Common DEGs were identified and their functional pathway enrichment analysis was performed by NetworkAnalyst. Cytoscape software was used for network visualization and hub gene identification. As a result of the analysis, several significant pathways were determined that possibly contribute to the shared etiology of AD and cancer. We also found five genes (CEBPD, DCN, DST, FHL1, SLIT3) as common DEG for five datasets. However, "EGFR" for AH-breast cancer, "SOX9" for AH-colorectal cancer, "THBS1" for AH-lung cancer, and "VEGFA" for AH-prostate cancer were identified as the most significant hub genes in network analysis. Our results provide promising in silico evidence for experimental studies that could be designed for drug repurposing, developmentand risk assessment purposesand further elucidate the relationship between AH and cancer.
