Abstract (EN)

Alzheimer's Disease (AD) is a non-reversible, progressive neurodegenerative disease. In the pathogenesis of the disease, amyloid-beta (Aβ) monomers are converted into toxic oligomers and then creates plaques. MicroRNAs (miRNAs) are essential for survival and neuronal function. Many miRNAs have been shown to be important in AD neuropathology. In this context, we are aimed to investigate the possible roles of miRNAs thought to be the targets of APP and β-secretase genes on expression levels of these genes by forming Alzheimer's model with sufficient Aβ42 expression in SH-SY5Y human neuroblastoma cells. For this purpose, SH-SY5Y and human fibroblasts cells which is negative control, are cultured under optimum conditions; Aβ42 protein treatment was applied.Qualitative and quantitative measurements of amyloid production in cells were performed by ELISA and MTT analyzes and congo red staining.Expression levels of the miRNAs associated with the disease and the genes associated with these miRNAs were quantitatively determined by Real-Time PCR. In conclusion, after disease modeling, miRNAs and genes associated with them were tried to be shown on the neuropathology of the disease and statistically significant changes were observed after application of Aβ42 expression levels of some miRNAs and genes related to the disease.
