Abstract (EN)

The aim of this study was to determine the functions of the AT3G23770 and AT4G14080 genes encoding ß-1,3 glucanase (callase) during callose degradation in the anthers of Arabidopsis thaliana (L.) Heynh. The seeds of T ? DNA knock-out mutants deficient in these genes were obtained from the ABRC Stock Center (Columbus) through Prof. Dr. Rod J. Scott (University of Bath) . İn this study, the stock number N859965 and CS850174 lines were used for AT3G23770 and AT4G14080. The seeds were germinated under labarotary conditions and mutant lines were analyzed phenotypically. These plants started to have flowers and siliques after 3-4 weeks of germination. The genotypes of these plants were determined using gen specific PCR. Based on the PCR results, homozygote (aa) mutant line for the N859965 and heterozygote (Bb) individuals for the CS850174 line were found. Also, male sterility and silique growth were analyzed on these plants. Afterwards, double mutants were obtained by crossing. Then, the genotypes of the hybrids were defined using PCR and homozygote plants (aabb) were selected. Flower, polen an silique growth of the double mutant lines were analyzed both cytologically and phenotypically, and the frequency of seeds per siliques was determined. Also, the expression profiles of the genes used in this project were investigated using RT-PCR.
