Protective effect of ß-Glucan on contrast induced-nephropathy in rats and comparison of its effect with nebivolol and N-acetylcystein

Abstract (EN)

One of the important mecanisms of the contrast-induced nephropathy (CIN) is the occuring hypoxia in renal medulla and oxidative stress. It was shown that ß-glucan (BG), which has antioxidant and immunomodulatory effects, attenuated the renal ischemia-reperfusion injury. We aimed to investigate whether BG might have a preventive role against the development of CIN and comparison of its effect with nebivolol and N-acetylcystein (NAC).Thirty-six Wistar-albino female rats were randomly divided into six groups (n=6 each): control, contrast media (CM), BG, BG+CM, Nebivolol+CM, NAC+CM. Exception of control and CM groups, the others were given their drugs once daily orally for 5 days. CIN was induced by administration of intravenous high-osmolal contrast media diatrizoate (6 ml/kg) after 48 h of dehydration. Kidney function parameters, inflammatory parameters, serum and renal tissue oxidative stress markers were measured. Kidney were excised for histopathological evaluation.Increases of serum creatinine and blood urea nitrogen levels in CM group only were significantly high (p<0.05). Absolute changes of serum creatinine levels in BG, BG+CM and Nebivolol+CM were significantly lower than those in CM (p<0.05). No significant decreases of creatinine clearance in all groups, except in CM and NAC+CM, were found. Regard to the absolute change of the creatinine clearance, differences between all groups were not significant. Serum levels of advanced oxidation protein product and malondialdehid in BG compared to CM were significantly less (p<0.05). Histopathological lesions in CM were more advanced (p<0.05). No significant differences between BC+CM, Nebivolol+CM and NAC+CM groups were found with regard to histopathologic findings.In conclusion, the study suggest that BG protects or ameliorates against CIN. Its beneficial effect may be similar to or more than those in nebivolol or NAC.Anahtar kelimeler: Beta glucan, contrast nephropathy, N-acetylcistein, nebivolol
