Investigation of Crisper Cas gene in Pseudomonas aeruginosaisolated from burn samples resistant to bacteriophage

Abstract (EN)

Burns are injuries to the skin or other tissues caused by thermal, radiational, chemical or electrical contact. Therefore, the patient is exposed to many microorganisms, including the bacterium Pseudomonas aeruginosa, which is known to be resistant to antibiotics. The aim of this study was to isolate Pseudomonas aeruginosa from burn patients and to determine the relationship between bacteriophage resistance of bacteria isolated from burns and the frequency of some Crispr-Cas genes for these bacteria. In this context, a total of 110 samples were taken from burn patients, including 25 men, 60 women and 25 children aged between 5 and 70 years. In 60 (54.55%) patients, the primary cause of burns was hot water, followed by hearth burns in 28 (25.4%) patients, oil burns in 15 (13.64%) patients and electrical burns in 5 (4.55%) patients. All of the present study samples were cultured on MacConkey agar medium and Cetrimide Agar. The cultured isolates were then subjected to various biochemical tests. 28 isolates were identified using Vitek-2 compact system and the rest were identified using Crispr-Cas genes. Female patients were mostly affected by Pseudomonas aeruginosa infection. Pseudomonas aeruginosa strains were found to be 100% resistant to all antibiotics except Colistin (75%). Genomic DNA extraction showed the presence of Cas5 in all samples analysed. It is thought that this may be related to antibiotic resistance
