Collagen extraction and characterization of skin and bone of some fish species

Abstract (EN)

This study aim of extraction and characterization of collagen from skin and bones of Scomber japonicus, Boops boops and Merluccius merluccius. Yield, moisture, crude ash and protein determination analyzes, amino acid analysis, Differential Scanning Calorimetry (DSC), Ultraviole and Visible Light Absorption Spectroscopy (UV-Vis), Fourier Transform Infrared Spectroscopy (FTIR) , X-Ray Diffraction (XRD) and Scanning Electron Microscopy (SEM) analyses were performed to determine the physicochemical properties of the extracted collagen samples. The collagen yield (%4.08-9.32) and protein contents (%91.00-93.60) of the skin samples of all species was higher than the collagen yields (%0.34-2.15) and protein contents (%87.35-90.20) (p<0.05) of the bone samples All collagen samples showed maximum absorption at 232 nm according to UV-Vis results. FTIR results support that the extracted collagen samples have triple helix structure. Thermal denaturation temperatures was determine range of 26.36-32.03ºC for skin samples and the range of 26.94-33.73ºC was found on bone samples. Fibrillar and porous samples was determine with SEM images. Glysin was the most abundant amino acid and there were relatively high contents of alanine, arginine, glutamic acid and aspartic acid. The present study found that collagen from fish processing wastes can be used as a high value material in many areas and most importantly, it is possible to prevent environmental pollution with evaluate fish processing waste.
