Kemal KOÇ: Prof.Dr.Ali ERKILIÇ: Yrd.Doç.Dr. Sema DÜZENLİIn this study, mature embrios of three wheat (Triticum aestivum L.) cultivarsgerminated under in vitro conditions and meristematic tissue parts of thesegerminated plants (mesocotyl) were cultured on MS medium containing differentconcantration and combination of different growth regulators.The best results wereobtained from 2,4-D, NAA and Dicamba (2.0 mg/l) and BAP (1 mg/l). Embryogeniccalli were obtained from mature embryo and two various media (variousconcentrations of 2,4-D with MS and 2,4-D + Dicamba with MS) were applied forinducing embriogenesis. The best results were obtained from 2.0 mg/l 2,4-Dconcentration and MS media to obtain embyiyogenik calli. As for the calliregeneration, various medias such as Glycine (1.0, 2.0, 3.0, 4.0 mg/l), BAP (1.0mg/l), Kinetin (1.0 mg/l) and IAA (0.1, 1.0 mg/l) were applied and the best resultswere obtained from including 0.1 mg/l IAA and 1.0 mg/l BAP plus MS media.Among the various Glycine concentrations (1.0, 2.0, 3.0 mg/l) the best results wereobtained from 1.0 mg/l Glycine concentration. The regenerated plants were found tobe the highest rooting from hormone free MS media. The rooted plants weretransferred into the soil and grown under greenhouse condition and the results showthat suitable and effective protocol were established for plant regeneration.Key Words : Triticum aestivum ,Mesocotyl, Mature Embrio, Calli, PlantRegeneration.II
