Abstract (EN)

Nowadays, studies on walnut tissue culture has gained grounds in Turkey, however it has not reached to the level that can meet the demands. In the present study, micro-propagation was carried out in order to produce walnut's rootstock and seedling in a rapid, clonal, and disease-free way. For this purpose and in order to sterilize explants obtained from auxiliary buds of specific walnuts (Vlach, Chandler, Yalova-1), the least contamination and a 70% actigen was applied for 5 minutes, was rinsed three times, each time the action of rinsing took 5 minutes. For shoot regeneration no difference was observed in terms of shoot regeneration in the three different walnut varieties used However, the different doses and combinations used in plant growth regulatory experiments, the maximum shoot formation per explant was 2,36 in MS medium containing 2 mg/L TDZ, 1 mg/L NAA, 0,2 mg/L GA3. To develop shoots' rooting procedure, the roots were supplemented with 4 mg / L of IBA in a modified MS medium in the Yalova-1 variety. The rooted shoots were treated with sterilized turf-perlite (1:1) mixture and conditioned for 28 days in a controlled conditions.
