Abstract (EN)
There are many methods used for chromosome detection in fish. These methods are mainly applied directly (in vivo) and indirectly (in vitro). Minimally invasive procedures are preferred in the application of the methods. However, in terms of the results to be obtained in this regard, the most appropriate method is in vitro studies, which are based on cell culture. Since the application of these high-quality chromosome-guaranteed culture techniques is long-term and much more costly, they are not routine methods for laboratories with limited physical and technological infrastructure. Although the direct methods, which are generally preferred in fish cytogenetic studies, are both practical and economical, applications such as keeping fish samples alive for a short time in vivo application of mitotic inhibitor treatment, euthanasia of samples under anesthesia with sedative agents, and incision of tissues according to the fish species limit the research. appearing as factors. Since the method of application of the mitotic inhibition pretreatment also requires adaptation to each fish species, it is also possible to waste time with preliminary trials to get good results. Especially in marine fish compared to freshwater fish, it is not always possible to keep the sample before and after the inhibitor if desired. For this purpose, studies to develop methods of obtaining chromosomes from a tissue piece taken from a living sample in a short-term in vitro manner are constantly carried out. In a very recent study, a much shorter and more profitable method based on tissue removal from dead samples, unlike other culture methods, has been developed. According to this method, which will also be applied in this study, the preparations prepared from the tissues taken in vitro after the death of the fish sample obtained from professional or amateur fishermen, which are known to have just come out of the sea, are stained with traditional Giemsa, and immediately followed by serial banding (AgNO3 staining and C-banding). It is considered to obtain high-quality chromosome sites in as short a time as direct methods. As a result of the study, it is aimed to reveal previously undetected chromosomal features of the genetic structure, which has an important role in the evolution of marine fish.
