Abstract (EN)
Phenolic compounds constitute a major group of organic contaminant. These compounds are used at a lot of industrial process, and are the degradation products of many natural compounds at the same time. At the low concentration levels, analysis of phenolic compounds produced at the result in fragmentation of polyphenols levels is very important in terms of figuring out their toxicity.In this thesis, apricot tissue, rich polyphenol oxidase enzyme, was used by immobilizing on oxygen probe and biosensor responds and detection limits to phenolic compounds were determined. In this study, optimization of bioactive layer component was carried out and 0.118 g/cm2 as apricot homogenate quantity, 6.64 mg/cm2 as gelatin quantity, and 2.5% as glutaraldehyde ratio were obtained. In the optimization studies, sodium-phosphate buffer (pH 8.5, 50 mM) and 37°C were determined as the optimum conditions. In addition, detection scale, thermal stability, operational stability, repeatability, substrate specify, interference effect and storage stability of biosensor were characterized. Detection limits of some phenolic compounds and polyphenols were measured and the highest biosensor responses were obtained from catechol and pyrogallol.With this study, it was reported that apricot tissue homogenate-based biosensor is utilizable for detection of various phenolic compounds.
