Abstract (EN)

In this research, antioxidant activities of six different active extracts obtained from Humulus lupulus L. plant were compared by DPPH, TEAC, FRAP and CUPRAC methods hexane, acetone following methanol, methanol (gradually) and methanol (directly) extracts were found as the most efficient. The amount of total phenolic components determined by F-C reagent, were found with the highest value for the mixture of ethanol 75% and water 25% extract. The enzyme activity/inhibition assays were performed by protease, tyrosinase and lactamase and % inhibition values of extracts were determined by calculating Vmax values. The cytotoxity effects of nine different concentration of active extracts were determined among Hep3B and HT29 cell lines by MTT assay. The number of living cells were determined by measuring absorbance at 570 nm. VLC was applied for the analysis of the n-hexane extract by varying mixtures of EtOAc and n-hexane. 20% EtOAc fraction were observed as the most active fraction by TLC and DPPH investigation of the fractions in MeOH. The structures of components obtained from methanol 100% fraction of SPE applied methanol extract were determined by spectroscopic methods. The components of crude extracts were determined by GC-MS/LC-MS. The purification and structural analysis studies of other active components are still continuing.Key words: Humulus lupulus L., Extracts, DPPH, CUPRAC, TEAC, FRAP, Cytotoxity, Enzyme activity/inhibition, VLC, HPLC-UV, GC-MS/LC-MS, Bioactive components.
