Identification of antioxidant, cytotoxic and enzyme active components from Lepidium sativum L. extracts

Abstract (EN)

Garden cress (Lepidium sativum L.) is an annual, fast growing herbaceous plant which belongs to Brassicaceae family. It has medicinal using since ancient ages. In this study antioxidant activity, enzyme-inhibition and cytotoxic activity studies for the extracts of aerial parts and seeds of garden cress were done. The antioxidant activity results for MeOH (fractional extracted) and MeOH (directly extracted) extracts are observed as the followings: IC50 values are 570 and 759 µg/mL by DPPH method, total phenolic contents are 47,62 and 28,30 mg ferulic acid/g extract by FC method, TEAC values are 5,26 ve 6,63 mM trolox/g extract by TEAC method, QERFC values are 0,55 and 0,72 by CUPRAC method, FRAP values are 0,48 and 0,43 mM FeSO4/mg extract by FRAP method. Protease, Tyrosinase and ß-Lactamase enzymes are used in enzyme-inhibition studies and it is observed that extracts did not inhibit the enzymes. Hep3B (hepatoma) and HT-29 (colon cancer) cells are used in cytotoxic activity studies and it is observed that extracts have toxic effect on Hep3B cells for certain concentration ranges and intensifier effect on existence of HT-29 cells. Seperation-purification and identification processes for bioactive components are proceeding according to the antioxidant activity.Keywords: Lepidium sativum L., extracts, bioactive components, antioxidant activity, enzyme-inhibition, cytotoxic activity, DPPH, FC, TEAC, CUPRAC, FRAP.
