Abstract (EN)

In this study, firstly, 1H-benzimidazole compound (1) was prepared by using o-phenylenediamine and formic acid. Then, as a result of the reaction of 1H-benzimidazole compound with ethyl-4-fluorobenzoate in the presence of anhydrous K2CO3, ethyl-4-(1H-benzimidazol-1-yl)benzoate compound (2) was obtained. As a result of the reaction of the synthesized ethyl-4-(1H-benzimidazol-1-yl)benzoate compound with hydrazine hydrate solution in absolute ethanol, 4-(1H-Benzimidazol-1-yl)benzohydrazide compound (3) was obtained. benzimidazole derivative compounds (4-21) containing hydrazide-hydrazone group were synthesized by the reaction of 4-(1H-Benzimidazol-1-yl)benzohydrazide compound with various aldehydes. The structure elucidation of the synthesized compounds were achieved by their elementel analysis, 1H-NMR, 13C-NMR and Mass (ESI +) spectral data. Anticancer activity of 18 obtained compounds (4-21) against HCT-116 colon cancer cell line was screened. In particular, compound 6 (IC50= 2.159±0.475 μM) bearing the (5-phenylthiophene-2-yl)methylene group and compound 17 (IC50=1.821±0.222 μM) bearing the ((3-bromobenzothiophen-2-yl)methylene) group had a better efficacy than the reference compound 5-fluorouracil. Compound 7 (IC50=0.251±0.015 μM) carrying ([2,2'-bitiophen]-5-yl-methylene) group showed a better activity compared to camptothecin (IC50=0.441±0.016 μM) and 5-fluorouracil, our other reference compound. Keywords: Anticancer activity, Benzimidazole, Hydrazide-hydrazone, Synthesis and structure elucidation.
