Abstract (EN)

1THE EFFECT OF VITAMINE E AND SELENIUM ON BLOOD AND TISSUELIPID PEROXIDATION AND SOME ANTIOXIDANT ENZYMES IN THE RATSEXPOSED TO BENZO(A)PYRENEMELTEM KIZILABSTRACTThe effects of vitamin E and selenium blood and tissues lipid peroxidation and someantioxidant enzymes levels in rats exposed to benzo(a)pyrene was investigated. For thispurpose, 100 Wistar Albino female rats were divided in 5 groups, as group 1 (Control), group2 (BaP), group 3 (BaP + vitamin E), group 4 (BaP + selenium), and group 5 (BaP + vitamin E+ selenium).Group 1: only plasebo isotonic sodium chloride,Group 2: one dose 10,08 mg BaP,Group 3: single dose of 10,08 mg BaP + 100 mg/kg vitamin E were injected in everyother day during the twelve week study period.Group 4: single dose of 10,08 mg BaP + 0,8 mg/kg selenium were injected in everyother day during the twelve week study period.Group 5: single dose of 10,08 mg BaP + 100 mg/kg vitamin E + 0,8 mg/kg seleniumwere injected in every other day during the twelve week study period.Plasma and liver lipid peroxidation (MDA) levels were significantly increased(p<0.001) because of the effects of BaP in the group 2. The lipid peroxidation level wassignificantly decreased in the groups 3 (p<0.01), 4 and 5 (p<0.001) related to effects ofvitamin E and selenium compared to group 2. Liver peroxidation level was significantlydecreased in the groups 3 and 5 (p<0.001) compared to group 2. Liver peroxidation level wassignificantly decreased in the group 5 (p<0.001) compared to group 4.Erythrocyte GSH, GSH-Px and catalase levels were significantly decreased (p< 0.001)in the group 2. The increase of erythrocyte GSH level was significant in the group 32(p<0.001), group 4 (p<0.05) and group 5 (p<0.01) compared to group 2. Erythrocyte GSH-Pxlevel was significantly increased (p<0.001) in the groups 3, 4 and 5, depending in the effect ofvitamin E and selenium, compared to group 2. Erythrocyte catalase level was significantlyincreased in the group 3 (p<0.05), compared to group 2.The liver GSH, GSH-Px and catalase levels were significantly decreased (p<0.001)because of the effect of BaP in the group 2. The increased liver GSH level was statisticallysignificant in the group 3 (p<0.01), group 4 and 5 (p<0.001), depending in the effect ofvitamin E and selenium effects, in the compared to group 2. The increased liver GSH-Px levelwas statistically significant in the group 3 and 5 (p<0.001) in the compared to group 2. LiverGSH-Px level was significantly increased in the group 5 (p<0.05), compared to group 4. Theincreased catalase levels in the liver were statiscically significant in the group 3 (p<0.001) andgroup 5 (p<0.05) compared to group 2.The cancer development was observed especially in lung, intestine and ovariumtissues in the group 2 related to effects of BaP. Cancer was not observed in the other groupstreated with vitamin E and selenium. The results of the current study showed thatadministration of BaP caused an increase in the levels of plasma and liver lipid peroxidationand decrease in the levels of antioxidant enzymes in the erythrocyte and liver. BaP can causethe cancer development. On the other hand, administration of vitamin E and seleniumdecreased the levels of plasma and liver MDA, while increasing the levels of erythrocyte andliver GSH, GSH-Px and catalase.It is concluded that; it was observed that vitamin E and selenium were effective inpreventing the negative effects of BaP on the lipid peroxidation, antioxidant enzymes and itscarcinogenic effects.Key words: BaP, Vitamin E, Selenium, Lipid peroxidation, Antioxidant enzymes.
