Abstract (EN)

The aim of this study is determining maximum germination temperature of spores of Coprinus cinereus and the temperature resistant protein structures by comparing protein combinations of mycel and fruit-bodies which grows up at normal and higher temperature. Spores were germinated three times seperately on CYM media in 24 hours at 37 oC and average germination percentage was calculated. All applications were repeated at 40, 41, 42, 43, 44 and 47 oC temperatures. According to the result of the growth value of 43 different and pure monokaryon mycel at 43 and 46 oC, one higher temperature resistant monokaryon was chosen and paired with each other and, paired the another monokaryon which is not resistant on higher temperatures. Thus these fruit-bodies and mycels were obtained. In addition, monokaryon mycels which resisted or not resisted at higher temperatures, were chosed and then obtained. All proteins of fruit-bodies and mycels were extracted. Proteins were analysed by using SDS-PAGE electrophoresis method for separating their molecular weights. Nearly at 41,0 oC, spore germination rate (19,53%) have been reached it?s maximum value. The profiles of monokaryon mycel proteins were showed nearly similar except number 38. It has been established that profiles of fruit-body and mycel proteins were different. Key Words: Basidiomycetes, Germination, SDS-PAGE, Fruit-body, Mycel
