Abstract (EN)

Eggplant (Solanum melongena L.,) is one of the most important vegetables in many countries. It has a long growing season, which makes it one of the vegetables more exposed to pests, disease attack, environmental stress and parasitic weeds. Orobanche spp, also known as broomrape, is one of the parasitic weeds that can cause great damage to eggplant yields. It is known that in order to germinate, the seeds of these parasites must be exposed to certain chemicals called strigolactones [SLs], which are released by the roots of the host. The CCD8 gene is involved in the synthesis of strigolactones and plays an important role in controlling growth and development. CRISPR-Cas9 was used to manipulate the SmCCD8 gene in eggplant (Solanum melongena L). The objective of the was study to create eggplants resistant to broomrape by knocking out or modifying the SmCCD8 gene. Yamula eggplant cultivar was used for transformation. Using Agrobacterium-mediated plant transformation method, gRNA and Cas9 were transferred to eggplant. Eggplant cotyledons were used as the plant material for Agrobacterium transformation. Successful editing of eggplant lines was confirmed by Sanger sequencing. The induced mutations were stably inherited in T1 and T2 progenies. Our results based on CRISPR-Cas9 technology open the way to the development of eggplant genotypes resistant to parasitic weeds.
