Investigation into the cytotoxic and apoptotic effects of Cu(II) flavonoid complex on lung cancer cells

Abstract (EN)
Lung cancer is one of the diseases that cause the most death in the world. Thelack of a complete solution in cancer treatment also enables new treatment searches. According to the studies conducted in recent years, studies on metal complexes are increasing gradually dueto the promising effects of metal compounds used in cancer treatment. In this thesis study, the anticancer activity of the newly synthesized Cu(II) flavonoid complex ([Cu (queH-1) Cl (phen)]. 2.5H2O (queH = quercetin, bpy = 2,2ꞌ – bipyridine) which synthesis and characterization performed was investigated in vitro. Cytotoxic activity of the Cu(II) flavonoid complex was evaluate in human lung cancer cell lines A549 cells. The effect of Cu(II) flavonoid complex on cell viability was analyzed by SRB and MTT viability tests. Cell viability was confirmed by themore sensitive and reliable ATP viability test. Annexin-V, PI (propidium iodide) and Hoechst triple fluorescence staining was perfor- med to determine the cell death modes (apoptosis/necrosis). Flow cytometry system (Annexin-V measurement) to determine the mechanism of cell death (apoptosis/necrosis /autophagy) responsible forits cytotoxic effects, caspase 3/7 activity, oxidative stress measurement, cell cycle analysis) and M30 antigen method were used. It was determined that it inhibits cell growth in individual cancer cells depending on thedose and time, and the complex induces apoptosis in cancer cells, causing a decrease in their migration abilities. The effectiveness of cisplatin used in the treatment of lung cancer and the newly synthesized Cu(II) flavonoid compound on cell viability was compared. As a result of the studies, it was concluded that further in vivo experiments should be carried out with the prediction that Cu(II) flavonoid complex could be used as a promising agent in the treatment of lung cancer.
