Abstract (EN)

ABSTRACT EVALUATION OF EFFECT OF ASPROSIN ON PAIN AND BEHAVIORAL FUNCTIONS IN MICE WITH EXPERIMENTAL MIGRAINE MODEL Migraine is a type of moderate to severe, usually throbbing headache that occurs periodically in attacks, often localized in one half of the head, and is often accompanied by nausea, vomiting, photophobia, phonophobia and osmophobia. The endocrine system, including adipose tissue, plays a vital role in pain perception and modulation. Dysregulation of the endocrine system has been associated with the development of various chronic pain conditions, emphasizing the importance of understanding the interaction between hormones and pain. In our study, we aimed to examine the effect of asprosine, an adipokine synthesized from adipose tissue, on pain and behavioral functions in mice with experimental migraine model. In this thesis, asprosine and sumatriptan were administered i.p. every other day for 9 days (days 1, 3, 5, 7 and 9) to mice in which an experimental migraine model was induced with nitroglycerin. Periorbital von-frey tests were performed on day 1 and day 9 to evaluate the mice's responses to pain, route-rod test on day 3 for motor coordination, open field test on day 5 for anxiety and depression, and object recognition test on day 7 for learning and memory. The groups of the study were as follows: healthy group, nitroglycerin-only group (control group), nitroglycerin plus asprosine group, and nitroglycerin plus sumatriptan group. When the results of the study were examined, a statistically significant decrease was found in the periorbital mechanical threshold value in mice with experimental migraine induced by nitroglycerin in the periorbital von-frey test performed on the 9th day, and in the group given sumatriptan, it was found that it reversed the effect of nitroglycerin in the periorbital von-frey test and statistically significantly increased the periorbital mechanical threshold value in mice. No statistically significant differences were observed in the other tests and groups compared in the study. In conclusion; we found that asprosine may increase the pain threshold and improve locomotor activity and behavioral functions, although not statistically significant in mice in which we created an experimental migraine model, but we think that the relationship between asprosine and pain and behavioral functions can be revealed more clearly in future more comprehensive experimental and clinical studies.
