Comparing the expression levels of mirna204-5P, mirna 138-5P and mirna 133-3P in formaline fixed – paraffine embedded tissue samples of endometrial hyperplasia and assesment of prognostic value

Abstract (EN)

The aim of our study is to determine and compare miRNA 204-5p, miRNA 138-5p and miRNA 133-3p expression levels in formaline fixed-paraffine embedded tissue samples of endometrial hyperplasia (EH) to assess their role in predicting and preventing endometrial cancer as a directive study in addition to other researches. This study includes 38 patients with atypical hyperplasia (1.patient group), 44 hyperplasia patients without atypia (2.patient group) and 44 patients with benign endometrial sampling (control group) admitted to our clinic between January 2005 – February 2015. Using qRT-PCR method, miRNA 204-5p, miRNA 138-5p and miRNA 133-3p expression levels of each group were evaluated. Kolmogorov Smirnov, Kruskal Wallis and Dunn tests were used for statistical analysis. A p value below 0,05 was accepted as significant. miRNA 204-5p expression levels were significantly lower in the EH patients comparing to control group (p=0,004 and 0,001, respectively). Comparing to control group, miRNA 138-5p and miRNA 133-3p expression levels were significantly higher in atypical EH group (p=0,005 and 0,001, respectively) whereas they were significantly lower in EH group without atypia (p=0.001 and 0,001, respectively). Increased expression levels of miRNA 138-5p and 133-3p in patients with atypical EH were in conflict with researches in literature. Clinical significance of this result remained unclear. This is the first study evaluating the levels of miRNA expression in FFPE samples of EH in such a large cohort consisting of 126 cases. Significantly lower miRNA 204-5p levels in EH cases gives an impression that it could be a potential biomarker used to predict malign transformation.
