Abstract (EN)
In recent years, diagnosis of several microorganisms and genetic diseases using Real Time PCR method could be performed with fully automated equipments.Real Time PCR test systems to detect genetic and microbial mutations are usually based on melting curve analysis which is not targetting to specific nucletide but only the random mutations. Besides this analysis system is not detecting the target mutation nucleotide, as if it looked to be able to detect the target mutations. Thus, this system could not be approved as a confirmatif test systems.Because of this disadvantages for melting curve analysis based on Real Time PCR test systems to detect single nucleotide mutations are not widely used. Furthermore, according to the literatures other methods except RFLP and DNA Sequencing are not reliable. For these reasons Real Time PCR methods based on/using TaqMan and Scorpion probs will be prominent in near future.Basically the test systems we will develop are going to use TaqMan and/or Scorpion primer-prob combinations. We belive that this combination will be able to detect the single nucleotide polimorphism we are targetting in manner of rapid, confirmative and reproducable.The Project described above will be aquired know-how in biotechnologies in the country via developments of new technologies .
