Characterization of hospital -acquired Pseudomonas aeruginosa strains with SDS-PAGE and PFGE methods

Abstract (EN)

P.aeruginosa is one of the most significant agent of hospital-acquired infections. Because of increased length hospitalization and cost of treatment , hospital infections are very important in developing and also in developed countries. Microbiologic diagnosis of P.aeruginosa infections is necessary for decreasing of hospital infections and developing an effective treatment plan.Because of high prevalance of P.aeruginosa infections, typing of strains with molecular methods is very important as well as isolation and identification of P.aeruginosa. Because of low reproducibility and discriminatory ability, phenotypic identification methods could not distinguish similar phenotypic strains with different genotypes.Recently, genotypic methods which analysed genom properties for phenotypic variations have been using for identifications. In this study, ıt?s aimed to investigate relationship of phenotypic and genotypic methods for identification of hospital-acquired P.aeruginosa strains. A total of 24 P. aeruginosa isolates which isolated from hospital-acquired infections over a ten- month period from November 2006 to September 2007 were included. Isolates were characterized by antibiotyping as determined by the disk diffusion method (Kirby-Bauer). PFGE and SDS-PAGE method were performed with isolated strains PFGE patterns obtained from twenty four pseudomonal strains were analysed. Typing of isolates were made according to DNA and protein profile. Of 24 P.aeruginosa isolates, 20 types were evaluated with antibiotyping, 13 type with SDS-PAGE and 20 type with PFGE methods. As a result, genotyping methods have more sensitivity because of reproducibility, discriminatory ability and allowing to typing at the base of genom properties.
