Abstract (EN)
In this study, aerobic endospor forming Bacilli group, isolated from the airborne environment in Seyitömer district of Kütahya province, were identified and some cabilities were investigated. Fifteen aerobic endospores producing bacteria were isolated by petri plate method based on gravity. Identification of isolated bacteria was carried out by biochemical tests at the genus level as Bacillus sp. When we look at the ability of enzyme production, it is determined that 6 isolates can produce amylase enzyme, while 14 isolates show protease activity. The highest MIC (Minimal Inhibition Concentration) value against heavy metals in comparison with reference strain (Bacillus subtilis NRRL B-209), by conducting the heavy metal resistance test, 93,33% of the bacilliies tested were found to be resistant to copper heavy metal. Antibiotic resistance testing was performed using agar disc diffusion method against eight different antibiotics (amphicillin, chloramphenicol, ciprofloxin, erythromycin, kanamycin, oxacillin, streptomycin, vancomycin). According to the test results, 100% of Bacilli were found to be resistant to oxacillin and 46,66% to ampicillin antibiotics. Supernatant fractions obtained from Bacillus isolates were tested against them self and other bacilli by agar drip method to determine metabolite production capabilities. S6 isolate activity was the best isolate in Bacilli group and S4 isolate was the most sensitive isolate. In addition, the antimicrobial activity of the supernatant obtained from the S6 isolate was tested against gram positive and negative microorganisms and yeast. S6 isolate showed the highest activity against Legionella serogroup 2-15. The effect of temperature, pH, organic solvent and detergents on the metabolic activity of S6 isolate was investigated. It has been determined that the S6 isolate is not active at temperatures of 80, 95 and 121°C in the supernatant, but is effected at different pH ranges (2, 4, 6, 8 ve 10) and in a small amount of organic solvents. Key Words: Antibiotic resistance, Bacillus sp., Enzyme, Heavy metal resistance, Metabolite.
