Abstract (EN)

Investigation that Antioxidant Activity of Various Extracts of Hippophae Rhamnoides and Juniperus Drupacea Aim: It was aimed to determine antioxidant activity of fruit parts of plants which are the Juniperus Drupacea grown in Mersin region and Hippophae Rhamnoides grown in Erzurum region and their effects on liver toxicity in rats. Material and Method: Radical scavenging, electron reducing power and metal chelating methods were used in antioxidan activity analysis. Total phenolic content quantitation were carried out by folin-ciocalteu reagent. In quantitative determination of α-tokoferol and retinol, HPLC- Uv-Visible and fluorescence detectors were used in rat liver samples. HPLC instrument. Gradient elution, methanol/acetonitrile/water mobil phase mixture, C18 column, 1 mL/min flow rate and 10 µL injection volume parameters were preffered for analysis. Results: UV-Görünür Bölge Spectrophotometer and HPLC methods developed after method validation tests were performed. Accuracy of the three methods (% BH) values of % 4.80 and precision (% RSD) values less than % 8.42. It was determined average analytical recovery value from tissue using UV detector and flourescence detector respectively, % 104.80 and % 92.467. HPLC method with UV, fluorescence detectors for tocopherol and retinol LOD values respectively, 0.220-0.053 and 0.025-0.050 µg/mL LOQ values were 0.712-0.150 and 0.100-0.257 µg/mL was detected. Conclusion: It was detected that Juniperus Drupacea and Hippophae Rhamnoides extracts show certain degree of antioxidant activity, particularly Juniperus Drupacea ether and Hippophae Rhamnoides water extract to be effective in liver damage. As antioxidants are known and found in liver tissue α-tocopherol, retinol in the blood of our appreciation of the qualitative and quantitative analysis can be done using HPLC-UV and fluorescence.
