Effect of different applications to increasing haploid embryo acquisition through gynogenesis in cucumber (Cucumis sativus L.)

Abstract (EN)
In this study, the effect of different applications was investigated to increase the yield of haploid embryos by gynogenesis in cucumber (Cucumis sativus L.). In the study, three different types of cucumbers (baby, mini, pickling) were used in 4 different media (C: CBM + 10/1 KINETIN:2,4-D + 3% sucrose + 0,7% agar, CS: CBM + 10/1 KINETIN). :2,4-D + 3% sucrose + 0,7% agar + 50mg/L spermidine, CP: CBM + 10/1 KINETIN:2,4-D + 3% sucrose + 0,7% agar + 50mg/L putrescine, CSP: CBM + 10/1 KINETIN:2,4-D + 3% sucrose + 0,7% agar + 50mg/L spermidine + 50 mg/L putrescine) were cultured. The cultured ovaries were kept in the dark at 35⁰C for 3 days and then at 24⁰C for 2 days. The cultured ovaries were finally kept in 24⁰C light for 9 days. The cultured ovaries were taken into the regeneration medium combined with MS + 4:1 (BAP:NAA) hormones after 2 weeks. After about 3 weeks, the embryos formed on the ovaries were recorded. All embryos formed were transferred to tubes containing hormone-free MS medium. Haploid plantlets were obtained from healthy embryos that completed their development. 309 embryos were obtained from 607 ovaries used in the experiment. The genotype that produced the most embryos was the genotype 1260. As a result, the medium that formed the most embryos was CP: CBM + 10/1 KINETIN:2,4-D 3% sucrose + 0,7% agar + 50mg/L putrescine medium and consisted of 126 embryos in total. A total of 106 plants were obtained from the resulting embryos. The maximum number of plants was obtained from CS: CBM + 10/1 KINETIN:2,4-D + 3% sucrose + 0,7% agar + 50mg/L spermidine medium. Genotype 1259 provided the highest plant yield. Embryo success status of the medium was compared according to genotypes. The genotype that produces the most embryos in the CP environment is pickling. mini cucumber genotype gave the highest number of embryos in C, CS and CSP media.
