Abstract (EN)
The rapid increase in the world population and the occurrence of climate changes are diminishing the quality and productivity of agricultural products. Issues such as nutritional disorders or inadequate nutrition arising from these factors make it imperative to thoroughly investigate the mechanisms developed by plants in response to stress conditions. The identification and characterization of genes involved in stress response, along with these mechanisms, are of great importance. Pathogenesis-related proteins are fundamental components of plant defense mechanisms. Studies have revealed that these proteins not only respond to biotic stress but also exhibit increased expression under abiotic stress conditions. However, the roles they play in response to abiotic stress factors have not yet been fully determined, leading to a significant gap in the literature. Based on this, in this thesis study, different vectors have been prepared to facilitate the determination of the heterologous expressions of SlPR-1.1 and SlPR-1.2 genes, reported to be upregulated under drought stress. While the cloning of genes into the pIPKb004 plant expression vector was conducted using the Gateway cloning method individually, the cloning of the two genes into the pGFPGUSPlus plant expression vector was carried out using traditional cloning methods. The presence of genes within the designated vectors was confirmed through PCR and restriction digestion reactions. The obtained plant expression vectors will enable the detection of stress responses, especially under abiotic stresses, resulting from the separate and combined expression of the two genes.
