Abstract (EN)
Breast cancer, which affects many women around the world, is an important health problem in the Middle East. The BRCA1 and BRCA2 genes, which produce proteins important for breast cancer that help suppress the growth of tumors in the body, increase the risk of developing breast cancer. In the study, chemical tests, PCR, and Sanger sequencing analyzes were applied to 8 patients. Hemogram data, BRCA1-ex10a amplification, and BRCA1-ex10b amplification results were found to be similar. In this study, seven SNPs (rs16941, rs799917, rs1799950, rs55906931, rs778359104, rs786203979, and rs2053655843) were found to show significant prevalence in Iraqi subjects with breast cancer. Each of the currently identified SNPs is shown separately to assess the nature of its possible interaction with the onset and development of breast cancer. Sequencing was done with 7 SNP Sanger arrays for 8 samples. This analysis was performed with the BRCA1 gene ID: 672. The rs16941 SNP identified in the BRCA1 gene has been extensively studied about breast cancer risk, and conflicting results have been reported in various populations. The rs1799950 SNP identified in the BRCA1 gene has been shown to have contradictory and complex associations with increased breast cancer risk. The detected BRCA1-rs55906931 SNP has a very low occurrence of the alternative G allele. The detected BRCA1-rs778359104 was found to be a rare faulty SNP located in the coding sequences of the BRCA1 gene, while the detected BRCA1-rs786203979 was reported to be a rare SNP located in the coding sequences of the BRCA1 gene with a false effect on the BRCA1 encoded protein. The rs2053655843 SNP, which was recently identified in this study, was found to be a rare SNP located in the coding sequences of the BRCA1 gene with a false effect on the BRCA1-encoded protein. Although the rs16941, rs799917, rs1799950, rs55906931, rs778359104, rs786203979, and rs2053655843 SNPs identified in the BRCA1 gene may be associated with an increased risk of developing breast cancer, the relationship between this SNP and breast cancer risk is complex and multifactorial.
