Abstract (EN)
This study was designed to synthesize AgNPs from supernatant-free cellcultures of some soil bacteria. Three bacteria (Pseudomonas aeruginosa (strain ALK320), Bacillus cereus (strain HNB5), and Corynebacterium striatum (strain 1910I14)) have been isolated and confirmedby PCR with universal primers. Then, the sequence results aligned with National Center for Biotechnology Information (NCBI) to ensure the species. These three bacteria were grown in nutrient broth to produce nanoparticles. Many approachesareusedtoverifytheproduction of AgNPs. Theystartwithcolorchanges, UV-visible, TEM, Scanning Electron Microscope, and end with EDX. The polymerase chain reaction (PCR) was used to identify Leishmania tropica (L. tropica); in which two primers were used to amplifythe 18sRNA gene, and they are a specific primer of gene LITSR/L5.8S, which gave an amplification product of 450bp, and a specific primer of gene mkDNA, which provided an amplification product of 120bp. The sequencing results showed a matchof 100% for L.tropica in the NCBI using the Basic Local Alignment Search Tool (BLAST). The maximum antileishmanial activity of extracellular AgNPs on L. tropica is recorded as growth inhibition (64.72%)at 400 μL /mL after being suppressed following 24 h of incubation by AgNPs synthesized using P. aeruginosa, B. Cereusand C. Striatumassays.The MTT assay revealed incredible results for AgNPs as Leishmania antiparasitic activity, to sum up, our novel approach to leishmaniasis treatment opens up new possibilities. Also, these AgNPs synthesized from natural materials can be used to treat a wide range of pathogenic bacteria and parasites.
