Determination of apomixis possibilty in Kaman walnut genotypes

Abstract (EN)

It is aimed to expose possibility of apomixis in Kaman walnut genotypes by molekuler markers in this research. The plant materials in this study were 40 Kaman-1 and Kaman-5 seedlings obtained from two nurserymen who produce them as a apomictic and 20 trees planted prevously as a apomictic that have partial nut production and orijinal Kaman-1 and Kaman-5 genotypes. Finally, 62 walnut plants were used in this project. SRAP and ISSR molekuler marker techniques were used in this study. Usage of SRAP marker was the first time in walnut and 10 primers produced totally 185 bands and 100 of them were polymorphic.while 15 ISSR primers amplified 200 bands and 122 of them were polymorphic. Genetic similarity coefficents among all Kaman-1 and Kaman-5 individuals changed between 0,60 and 0,93. As a result, this study demonstrated that non-grafted walnut seedlings sold as a apomictic in Kaman are not apomixis. In addition , ISSR markers differentiated walnut seedlings better than SRAP ones. Furthermore, the correlation between ISSR and SRAP genetic similarity matrices was foud very low (0,22). Key Words : Walnut, SRAP, ISSR, Genetic diversity, Juglans regia.
