Abstract (EN)
Carboxidotrophic bacteria offer numerous opportunities in biotechnological processes due to their ability to tolerate and grow in toxic CO gas. Carboxydotrophs carry out the oxidation of CO through the enzyme CO dehydrogenase (CODH) and converting it into less harmful compounds. These bacteria, capable of oxidizing CO, can be regarded as a potential resource in various areas such as environmental remediation, bioenergy production, and industrial applications. In this study, water samples taken from acid mine drainage were cultured for the isolation of carboxidotrophic bacteria. As a result of the isolation, Bacillus subtilis and Bacillus sp. identified, carboxidotrophic bacteria could not be isolated. After isolation it was aimed to amplify this gene region in Pseudomonas aeruginosa, where DNA sequences encoding the synthesis of aerobic carbon monoxide dehydrogenase (CODH) enzyme are seen in the genomic sequence. By examining the genomes of Pseudomonas aeruginosa B136-33 and Pseudomonas aeruginosa ATCC-27583 strains, target gene regions for amplification were identified. Primer design and PCR amplification processes were completed. Based on the conducted experiments, it can be concluded that the Pseudomonas aeruginosa strains in the laboratory do not possess the CO dehydrogenase sequence found in aerobic carboxydotrophic microorganisms. In future studies, different marker genes could be selected and used for the intended research.
