Abstract (EN)

In the present study, carboxymethyl cellulase, xylanase, and lichenase producing bacterium Bacillus sp. CS8 was isolated from the soil samples collected from agricultural greenhouse located in Adana province. The optimum activities were observed at 40 °C for all three enzymes. On the other hand, optimum pH values were 7.0 for CMCase and lichenase, whereas 6.0 for xylanase. CMCase, xylanase and lichenase showed 57, 83, and 71% residual activity after pre-incubation at 80 °C for 15 min. The relative residual activities between 50-80 °C occured as 80.25, 87.0, and 89.25%, in the same order. Maximum CMCase and xylanase productions of the isolate were observed after 36 hours following the inoculation time, whereas 48 hours after for lichenase. All three enzyme activities were stimulated by CaCl2, MgCl2, and MnCl2, whereas inhibited by HgCl2 and CuCl2. While ZnCl2 stimulated the CMCase and xylanase activities, FeCl3 stimulated CMCase and lichenase, and EDTA stimulated xylanase and lichenase activities. When xylanase activity was assumed as 100%, the activities of CMCase and lichenase in comparison to the xylanase remained as 89 and 79%, respectively.
