Abstract (EN)

In the present study, dominant AFLP method and its derivatives such as SAMPL and TE-AFLP techniques with a codominant SSR method were used to determine the most polymorphic primer combinations and the most polymorphic marker system in self-pollinated monoecious P. atlantica population to use for genetic-mapping studies. One monoecious P. atlantica and its ten progenies were used. In AFLP method, 96 EcoRI-MseI primer combinations generated a total of 8177 fragments that 603 were polymorphic. Total of 358 polymorphic SAMPL markers were produced by 96 SAMPL/MseI primer combinations. In TE-AFLP method, 8 Xba/BamHI primer combinations produced 302 fragments which 36 were polymorphic and in SSRs, 33 primer combinations generated 50 SSR allelles that 18 were polymorphic. The results of this study showed that the most polymorfic marker system is AFLP and AFLP method can be used with codominant SSR method together for genetic-mapping studies.
