Abstract (EN)

Target-based manipulations of the stem cell fate with small chemical molecules both provide new insights into stem cell biology and contribute to the development of therapeutic approaches with stem cells. Small chemical molecules are regulators that target cell signals and mechanisms and play a role in stem cell fate. This thesis, it is aimed to increase the efficiency of traditional methods used in the differentiation of rat bone marrow-derived cells and adipocyte cells. In this study, small chemical molecules that can be effective in directing the differentiation of rat bone marrow-derived stem cells into adipocytes were applied to the cells in vitro. In the next step, adipocytes specific gene expression profiles were analyzed. When the results were examined, an increase was observed in the gene expression of genes that are important in adipocyte differentiation of the cells treated with Forskolin, CHIR99021, SGC0946 chemicals compared to the untreated negative control group. It is thought that the obtained data can be used as a therapeutic agent for regenerative medicine, if some complementary studies are conducted to better understand the subject of the study and if the yield is effective in different species in differentiation to adipocytes.
