A research on in vitro clonal propagation of Kitaibelia balansae

Abstract (EN)

Kitaibelia balansae is from perennial herbs which is endemic to Cilicia and Levant and classified as `Endangered? in territory of Turkey. Only one sample stalk with roots of Kitaibelia balansae was found in Gaziantep vicinity and transplanted to Botanic Garden of Gaziantep. In this research, for biodiversity conservation we conducted to determine for vegetative propagation potentials from leaves and petioles of Kitaibelia balansae throughout in vitro tissue culture methodology. Plant samples from introduced to Botanical Garden of Gaziantep were collected, after surface sterilization explants leaf blade with midrib and petioles were subcultured on basal MS medium supplying with same hormones at various combination and concentration 6-Benzylaminopurine (BAP), Kinetin, Indole-3-Acetic Acid (IAA), 1-Naphthaleneacetic acid (NAA), Gibberellic Acid -3 (GA3), Indole-3-butyric acid (IBA) and 2,4-dichlorophenoxyacetic acid (2,4-D) suplemented with 3 % sucrose and 5 %agar as solidifying agent. Then they were incubated in the climate room at 25±1 ºC temperature with 16 hours fluorescent light for 4 months. Callus induction freguency at different levels was observed in two months on basal MS including all hormone varieties treated in the experiment. Homever embryogenic green callus were significantly induced on medium containing.0.5 mg/l Kin + 0.5 mg/l IBA and 0.5 mg/l Kin + 0.01 mg/l 2,4-D in 15-20 days. Then they turned to brownish in approximately two months, nevermore the results of four months laboratory experiment seem very promising for clonal multiplication from embryogenic callus of Kitaibelia balansae by tissue culture.Key words: Kitaibelia balansae, tissue culture, callus, propagation
